Abstract
The current study was conducted in Wasit Province / Iraq and included 96 stool and 96 female urine samples from suspected
patients of parasitic infection caused by Enterobius vermicularis during the period from October 2019 to April 2020. All
samples were examined using direct wet smear and Luoglos iodine stain , Scotch method and then cultivated the samples on
blood and MacConkey agar for diagnosing using API 20E, API staph, and polymerase chain reaction (conventional PCR).
The results of the stool sample showed that 54 positive samples (56.25%) out of 96 patients when examined by Direct &
Luoglos iodine and 16 positive samples (84.21%) out of 19 patients when examined by Scotch tape where the results showed
35 positive samples (36.4%) of the total 96 a result in the studied samples by PCR method. Regarding the conventional PCR
method was performed to detect E. vermicularis based on the 18S ribosomal RNA gene; as for urine culture samples, the
results were 50 positive samples (52.08 %) out of 96 belonging to different types of bacteria. After this, a PCR examination
was conducted for 56 samples of the same stool samples to verify the presence of bacteria, and the results came 24 (42.85%)
for the types of bacteria that were culture on blood agar and MacConkey agar . During the present study, it was found that
the incidence rate in the rural areas is more than 53 (50.88%) in the urban areas 43 (41.28%).

Key words: PCR, Scotch tape, Enterobius vermicularis, Bacteria, Culture
