THYROID GLAND AND BREAST CANCER

Abstract
The link between thyroid gland disorders and breast cancer was intensively debated for years. Some breast
diseases may be considered as iodine deficiency diseases.Over-expression of NIS in breast cancer samples was
reported. Additionally, it was found to be positively associated with ER-α expression. Recently, a role of iodine
was suggested as an ER-α agonist thus could mediate the growth of breast cancer cells. A higher frequency of
TPO Ab, was found in patients with BC. BC patients with increased TPO Abs tend to have a favorable outcome.
Many mechanisms were proposed to explain this relation however none of them is universally agreed.
Hypothyroidism was initially considered as a poor prognostic factor for BC. Conversely, an increased risk of BC
was described in patients affected with hyperthyroidism. Thyroid hormone receptors could become a marker and
a therapeutic target like the estrogen and progesterone receptors. High THRA expression and disease-free
survival was noted in invasive breast cancer. Additionally, specific alterations in TR-α and TR-β gene expression
was demonstrated in breast cancer. TRs mediate multiple effects of T3 on the phenotype, proliferation and gene
expression of cultured normal mammary epithelial cells .it was observed that the expression of these receptors
suffers important modifications in the different pathologies Thus, only normal breast showed nuclear
immunoreaction for TR-α and this location was changed in BC suggesting a deregulation of its target genes
which could trigger breast cancer development thus these receptors may serve as new therapeutic targets for BC.
