ANDROGRAPHIS PANICULATA ATTENUATES DIETHYLHEXYL PHTHALATE-INDUCED DETRIMENTAL CHANGES IN GLUCOSE OXIDATION AND INSULIN SIGNALING IN HUMAN CHANG LIVER CELLS

Abstract
Aim: The present study examined the effects of Andrographis paniculata in diethylhexyl phthalate (DEHP) exposed
Chang liver cells in vtro. Methods: Chang liver cells (5x105
cells) were exposed to 400 M of DEHP for 24h. After
different concentrations A.paniculata powder extract for 48 h. After 48h, the cells were used for the assessment of
insulin receptor (IR), insulin receptor substrate-2 (IRS-2) mRNA and glucose oxidation. Results: IR and IRS-2
mRNA and glucose oxidation in DEHP exposed Chang liver cells were significantly reduced (p<0.05). Cells treated
with A.paniculata extract enhanced IR and IRS-2 mRNA and oxidation of glucose in a dose-dependent fashion.
(p<0.05). Conclusion: The present findings are the first to report in identifying the antidiabetic activity of
A.paniculata against DEHP exposed detrimental changes insulin signaling molecules and glucose oxidation. Hence,
A.paniculata may have influence in controlling diabetes and hence it could be used a therapeutic herbal drug for the
treatment of diabetes mellitus. Further studies on the possible role of A.paniculata on the protein expression of
downstream insulin signaling molecules in liver cells are warranted in order to strengthen the potential of
A.paniculata.
